Alright, guys, I've been thinking it over and I have a general idea of what we're going to need. At the very least, we're probably going to need:
-An opening theme, probably based around Layton's normal theme -A puzzles theme -A theme for the main villain -A sort of walkabout theme, like London Streets -Something for suspicion/rising action -Climax/battle music -Sad refrain, like Iris or Time Travel, for the tragic parts -End credits
We won't have to do all of this from scratch, considering the volume of music we have to work with, but a good portion of it will have to be original. Any ideas?

Hi. I can write. I can play clarinet and I don't know if I can sing, but I don't think so.

I think we could just take the original puzzle theme and modify it slightly. I don't mean steal it, but like what they do in his other games. They kind of modified the original.
